I have an Apple Display 27 with mini display output, and I was wondering if I could replace the cable with a thunderbolt ! Is this possible ?

Sorry no! MiniDisplay signaling is different than Thunderbolt. You need to use the correct cable otherwise your display won’t work correctly.
